Colon Cancer Tissue Microarray Now Available
The Cancer Diagnosis Program is pleased to announce the availability of a Colon Cancer Tissue Microarray that contains 366 primary colon cancers along with appropriate diverticulitis, adenomatous polyps and normal colon mucosa as controls. Designed by a biostatistician, it is sufficiently robust to allow for critical testing of a marker’s prevalence in colon carcinoma as well as its association with clinical outcome. The accompanying brochure describes the clinical annotation documented in the TMA. Each tissue microarray
(TMA) set consists of
-
invasive colon carcinoma
- normal colon epithelium
- adjacent to the cancer: N=34
- diverticulitis, no cancer: N=40
-
adenomatous polyps
- controls
-
normal non-colon
tissues: N=4
- cell lines: N=6
The TMA was designed by NCI statisticians for high statistical power for comparisons
of marker values
- between colon cancers of different stages, or
- between colon cancers that recurred within a few years of diagnosis versus colon
cancers that remained recurrence free. For example, there is 80% power to detect
- difference in prevalence
rate for a binary marker
of about 0.30 between
Stage I and
Stage IV cancer
- difference in prevalence
rate of about 0.35
between Stage II colon
cancers that recurred
within 5 years versus
those that did not recur
within 5 years.
Each array is created in
quadruplicate to address
possible tumor
heterogeneity.
- Four cores are taken
from each case block,
with one of those cores
placed in each of four
replicate array blocks.

Information for invasive
colon carcinoma cases:
-
race, gender and age
at diagnosis
- year of diagnosis
- primary site within colon
- TNM stage (AJCC 5th edition)
- number of positive nodes and total number examined
- initial treatment— only Y/N: surgery, chemotherapy, radiation
- histology, grade, lymphovascular invasion, status of margins
- recurrence and time to recurrence
- vital status
- cancer status at last follow-up
Information for adenomatous polyp cases:

gender
- age at diagnosis
- year of diagnosis
- polyp type
- degree of dysplasia
- gross size of polyp
